I science fiction-filmen 'Cloud Atlas' kædes seks historier sammen mere eller mindre sammen. På en stillehavsø i 1850 møder notaren Adam Ewing (Jim Sturgess) en slave og en excentrisk doktor. I Belgien i 1831 stifter den unge Robert Frobisher (Ben Whishaw) bekendtskab med en kendt komponist og indleder et forhold til hans kone. I 1975 kommer reporteren Luisa Rey (Halle Berry) på sporet af en atomkatastrofe i Californien takket været en vigtig kilde. I det nuværende England udsættes forlagschefen Timothy Cavendish (Jim Broadbent) for en række absurde hændelser. I fremtiden udspørges klonen Sonmi~451 (Doona Bae) om sine skæbnesvangre handlinger, og i en endnu fjernere, dystopisk fremtid fortæller den gamle mand Zachry (Tom Hanks) om sine skelsættende oplevelser som ung.
